產(chǎn)品介紹
Norgen Biotek是來自加拿大的著名生物技術(shù)公司，生產(chǎn)基于技術(shù)上的核酸和蛋白質(zhì)純化及富集產(chǎn)品。Norgen Biotek 在2003年獲得加拿大國家研究委員會頒發(fā)的科技創(chuàng)新獎。目前該公司的產(chǎn)品多達(dá)40余種，覆蓋基因組和蛋白組，廣泛地應(yīng)用于不同的領(lǐng)域。
提純RNA的方法一般有兩種，離心法和有機(jī)抽取法（例如Trizol）。Norgen Biotek是世界上*家真正意義上的生產(chǎn)以離心柱（Spin Columns）為平臺，并能純化所有種類RNA（包括小分子RNA）的公司。
Norgen Biotek和其他公司生產(chǎn)的以離心純化為原理的試劑盒比：
- 其他公司一般都使用硅材料（silica matrix）為填充物。硅材料的缺點(diǎn)是在沒有酚存在的情況下，不能回收所有的RNA，使得其純化效率很低，特別是對小RNA（例如microRNA）,其回收率幾乎為零。這也是為什么很多客戶在使用了其他公司的以離心為原理的總RNA純化產(chǎn)品后，往往得不到預(yù)想結(jié)果的原因。
- 即使對少量細(xì)胞的總RNA提純，也具有很高的線性（linearity on low numbers of cells）。在細(xì)胞數(shù)量多的情況下，其他公司的產(chǎn)品表現(xiàn)還可以，但是在細(xì)胞數(shù)量少的情況下，其純化效率會大大降低，為提高效率往往需要通過其他的途徑，例如Carrier RNA。Norgen的試劑盒可以從一個細(xì)胞中提純出高質(zhì)量的總RNA，在同行業(yè)中處于地位。

Norgen Biotek和其他公司生產(chǎn)的以Trizol原理的試劑盒比較：
- 在這些方法中，層析會造成有機(jī)試劑的殘留，從而抑制許多下游實(shí)驗(yàn)的進(jìn)行。
- 許多實(shí)驗(yàn)室由于健康和環(huán)境的因素，已經(jīng)放棄了以酚抽為原理的提純方法。
- 對大樣品，使用Trizol的效果還可以，但同時(shí)也由于基因組DNA的增加，會阻止RNA的沉淀，從而間接地降低了純化效率
- 對小量樣品，使用Trizol的效果往往不佳。
- 使用Trizol中，乙醇沉淀（ethanol precipitation）效果的好壞和RNA的量有密切的關(guān)系，因此對于少量樣品，沉淀法往往得不到zui終的RNA產(chǎn)物。
- 由于許多實(shí)驗(yàn)（e.g. qRT-PCR），并不需要很多量的RNA，因此Trizol方法已經(jīng)逐漸被其他方法所代替。
Norgen Biotek采用的柱子填充物是一種新型的，并有保護(hù)的非硅膠材料?？梢栽跊]有酚等有害有機(jī)溶劑存在的情況下，純化包括microRNA在內(nèi)的所有RNA,而且其純化回收效率高達(dá)99%。人們一直認(rèn)為如果要純化microRNA,必須要用Trizol，而不是離心法。這種論斷已經(jīng)過時(shí)。Norgen Biotek總RNA純化試劑盒不僅避免Trizol帶給實(shí)驗(yàn)者的健康損害，而且可以純化出包括miroRNA在內(nèi)的所有RNA。因此Norgen Biotek是一款真正意義上的綠色產(chǎn)品。
