Chemicals and reagents

Dex was obtained from Shuanghe Pharmaceutical Company (Wuhan, China). RYR1 agonist 4-chloro-3-ehylphenol (4-CEP) was purchased from Macklin (Shanghai, China), and RYR1 inhibitor dantrolene (Dan), ERS agonist tunicamycin (Ti), ERS inhibitor 4-phenylbutyric acid (4-PBA) and Fura-2 AM calcium fluorescent probe were purchased from Meilunbio Biotech (Dalian, China). Fluo-3 AM calcium fluorescent probe was purchased from Beyotime (Shanghai, China).DMEM F12 medium was purchased from Gibco Co., Ltd. (Grand Island,NY, USA). Safranin O and fast green was obtained from Hengyuan Biotech (Shanghai, China). TRIZOL was purchased from Invitrogen (Carlsbad, CA, USA). A reverse transcription and real-time quantitative polymerase chain reaction (RT-qPCR) kit was purchased from Takara Biotechnology Co., Ltd. (Dalian, China). SYBR Green dye was obtained from Applied Biosystems, by Thermo Fisher Scientific (ABI, Foster City,CA, USA). All primers were synthesized by Sangon Biotech (Shanghai,China). Primary antibodies for RYR1 and 78 kDa glucose-regulated protein (GRP78) came from Sigma (St. Louis, MO, USA). The antibody for phosphorylated eukaryotic initiation factor 2A (p-EIF2A) was acquired from CST (Boston, MA, USA), the antibody for ATF-6 was acquired from Santa Cruz (Santa Cruz, CA, USA), the antibody for COL2A1 was acquired from Abbkine (Santa Cruz, CA, USA) and the antibody for ACAN was acquired from Bioss (Beijing, China). TGFβR1 antibody was purchased from ELK Biotechnology (Wuhan, China), and the antibodies for SMAD family member 2 (SMAD2), sex-determining region Y-box 9 protein (SOX9), ADAM metallopeptidase with thrombospondin type 1 motif 5 (ADAMTS5), and matrix metallopeptidase 13 (MMP13) were acquired from abcam (Cambridge, UK). The other reagents for the experiments were of analytical grade.
