CD28 是 T 細(xì)胞表面的核心共刺激受體,屬于免疫球蛋白超家族(IgSF),其通過與 抗原呈遞細(xì)胞(APC)上的配體 CD80(B7-1)和 CD86(B7-2)結(jié)合,提供 T 細(xì)胞激活的 “第二信號"(共刺激信號),是適應(yīng)性免疫應(yīng)答的關(guān)鍵調(diào)控節(jié)點。CD28 信號缺失會導(dǎo) 致 T 細(xì)胞失能(Anergy),而過度激活則與自身免疫病及移植物抗宿主?。℅VHD)相關(guān)。 CD28 共刺激信號的恢復(fù)可增強 CAR-T 細(xì)胞或 PD-1 抑制劑療效,成 為免疫調(diào)控的重要靶點。 CD28 激活對 T 細(xì)胞的共刺激會啟動信號級聯(lián)反應(yīng),導(dǎo)致 AP-1 和 NFκB 轉(zhuǎn)錄因子激活 和核轉(zhuǎn)位。這些途徑顯著增強 T 細(xì)胞細(xì)胞因子的產(chǎn)生,特別是 IL-2,從而促進 T 細(xì)胞增殖、 分化和存活。 | |
